Synthetic peptide IGF-1 LR3 is lengthened analogue of endogenous peptide hormone IGF-1 (human insulin-like growth factor 1). Unlike regular IGF-1, its prolonged version IGF-1 LR3 haves an arginine instead of a glutamic acid at the third position in its amino acid sequence (“arginine 3”), and also has an additional 13 amino acids at its N-terminus (MFPAMPLLSLFVN) (“long”). Total length of its amino acid chain is 83 amino acids unlike 70 amino acids of regular IGF-1.

IGF-1 LR3 still retains its strong pharmacological activity as an agonist of the IGF-1 receptor, but due to the mentioned modifications has very low affinity for the insulin-like growth factor-binding proteins (IGFBPs) unlike regular IGF-1 does have, and also has improved metabolic stability. As a result, it is approximately three times more potent than IGF-1, and possesses a significantly longer half-life of about 20 hours.
